1、A novel role for thyroid-stimulating hormone: Up-regulation of hepatic 3-hydroxy-3-methyl-glutaryl-coenzyme a reductase expression through the cyclic adenosine monophosphate/protein kinase A/cyclic adenosine monophosphate-responsive element binding protei

摘要:Elevated thyroid-stimulating hormone (TSH) and hypercholesterolemia commonly coexist, as typically seen in hypothyroidism, but there is no known mechanism directly linking the two. Here, we demonstrated that in liver cells, TSH promoted the expression of 3-hydroxy-3-methyl-glutaryl coenzyme A reductase (HMGCR), a rate-limiting enzyme in cholesterol synthesis, by acting on the TSH receptor in hepatocyte membranes and stimulating the cyclic adenosine monophosphate / protein kinase A / cyclic adenosine monophosphate-responsive element binding protein (cAMP/PKA/CREB) signaling system. In thyroidectomized rats, the production of endogenous thyroid hormone was eliminated and endogenous TSH was suppressed through pituitary suppression with constant administration of exogenous thyroid hormone, and hepatic HMGCR expression was increased by administration of exogenous TSH. These results suggested that TSH could up-regulate hepatic HMGCR expression, which indicated a potential mechanism for hypercholesterolemia involving direct action of TSH on the liver. 2、Novel inlet-airframe integration methodology for hypersonic waverider vehicle

摘要:With the aim of integrating a ramjet or scramjet with an airframe, a novel inlet-airframe integration methodology for the hypersonic waverider vehicle is proposed. For this newly proposed design concept and for the specified flight conditions, not only the forebody of the vehicle but also its engine cowl and wings can ride on the bow shock wave, causing the bow shock wave to remain attached to the leading edge for the entire length of the vehicle. Thus, this integrated vehicle can take full advantage of the waverider׳s high lift-to-drag ratio characteristics and the ideal pre-compression surface for the engine. In this work, a novel inlet-airframe integrated axisymmetric basic flow model that accounts for both external and internal flows is first designed using the method of characteristics and the streamline tracing technique. Subsequently, the design of the inlet-airframe integrated waverider vehicle is generated from the integrated axisymmetric basic flow model using the streamline tracing technique. Then, the design methodologies of both the integrated axisymmetric basic flow model and the integrated waverider vehicle are verified by a computational numerical method. Finally, the viscous effects and performance of both the integrated axisymmetric basic flow model and the integrated waverider vehicle are analysed under the design condition using the numerical computation. The obtained results show that the proposed approach is effective in designing the integrated hypersonic waverider vehicle.

3、From ZnSn(OH)6 to SnS2: Topotactic transformation synthesis of SnS2 hierarchical microcubes with superior Li-ion storage performance

摘要:Display Omitted• Uniform 3D hierarchical SnS2 microcubes were synthesized via a topotactic transformation route. • The anode of these 3D SnS2 microcubes exhibited stable cycling performance. • The anode of these 3D SnS2 microcubes also showed superior rate capability.The 3D hierarchical microstructured electrodes by assembling the individual 1D/2D nanosubunits can fully take advantage of the merit of 1D/2D nanostructured materials, as well as alleviate their drawbacks. In this work, SnS2 3D hierarchical microcubes consisting of 2D nanosheet subunits were facilely synthesized via a simple topotactic transformation with room-temperature coprecipitated ZnSn(OH)6 microcubes as the template. Compared to bulk counterparts, this microstructured SnS2 was found to have a unique morphology and electrochemical interfacial regions that facilitated highly efficient Li-ion insertion/extraction and effectively accommodate the large volume changes during charge/discharge cycles. Over the whole 200 cycles at 1C current density, an outstanding cycleing stability is exhibited, with a high stable specific capacity of about 640 mA h g−1.

4、Identification of pentacyclic triterpenes derivatives as potent inhibitors against glycogen phosphorylase based on 3D-QSAR studie

摘要:Naturally occurring pentacyclic triterpenes (PT), a novel class of inhibitors against glycogen phosphorylase (GP), hold promise for the treatment of type-2 diabetes and other diseases with disorders in glycogen metabolism. To identify novel and more potent GP inhibitors, the receptor-based comparative molecular field analysis (CoMFA) and comparative molecular similarity analysis (CoMSIA) approaches were performed to investigate the quantitative structure-activity relationships (QSAR) among 106 PT analogues. The validated models demonstrated that the elongated or bulky substitutions in C17 position and/or C2, C3 positions are favorable. Then based on the structural information extracted from these models, 56 derivatives were synthesized and biochemically tested in this study. The IC50 value of the most potent compound P50 was found to be 1.1μM.

6、Preparation and RGB upconversion optic properties of transparent anti-counterfeiting films1 2017

摘要:1 online resource Abstract : A series of anti-counterfeiting labels based on RGB upconversion inks was designed for the die-cutting packages, suggesting a highly advanced security effect for the package anti-counterfeiting application. Abstract : Advanced anti-counterfeiting labels have aroused an intensive interest in packaging industry to avoid the serious issue of counterfeit. However, the preparation and cost of the existing labels associated with the drawbacks, including the complex and high-cost equipment, limit the protection of the authenticity of goods. Herein, we developed a series of anti-counterfeiting labels based on multicolor upconversion micro-particles (UCMPs) inks via straightforward and low-cost solutions, including spin-coating, stamping and screen printing. The UCMPs were synthesized through a facile hydrothermal process and displayed tunable red (R), green (G) and blue (B) color by doping different lanthanide ions, which are Er 3+ /Tm 3+, Yb 3+ /Er 3+ and Yb 3+ /Tm 3+ in NaYF4 hosts, respectively. The optimal UCMPs inks were deposited on a flexible polyethylene terephthalate (PET) substrate to obtain transparent anti-counterfeiting labels possessing higher transmittance, stronger upconversion fluorescence intensity and good photostability. Under ambient conditions, the patterns and films were transparent, but could exhibit multicolor light under 980 nm laser excitation. They can be used as anti-counterfeiting labels for die-cutting packages to further elevate the security of goods. The tunable and designable transparent anti-counterfeiting labels based on RGB UCMPs inks exhibit the merits of low-cost, easy-manufacture and versatility, underlying the practical application in the field of anti-counterfeiting.

7、Dose-dependent target diversion of Danhong injection on the Glu-GLT-1/Gly-GlyRα dynamic balance module of cerebral ischemia

摘要:Function-oriented modular structure analysis is a great challenge in module-based pharmacological studies. A strategy to uncover target-target interaction (TTI) and dynamic balance regularity (DBR) was established to discover the structural factors influencing modular functions and explore the mechanism of Danhong injection (DHI) in treating cerebral ischemia. The dose-related metabolic features of DHI intervention were investigated using metabolomics and modular pharmacology. The findings indicated that Glu/Gly was a biomarker and Glu-GLT-1/Gly-GlyRα was the core unit regulated by DHI. Gly and Glu displayed opposite patterns and functional roles, representing intra-modular balance. GlyRα was identified as the upstream target and GLT-1 as the downstream target by inhibiting or activating GlyRα, indicating that DHI has two dose-dependent regulatory modes. GlyRα was the major target at low doses, while GLT-1 was activated as the dominant target as doses accumulated. Our study reveals that target-target interaction and dynamic balance regularity are the key factors influencing modular functions, which is a promising breakthrough for module-based pharmacological studies.

9、Paternal ethanol exposure and behavioral abnormities in offspring: Associated alterations in imprinted gene methylation

摘要:Research confirms that maternal ethanol (EtOH) exposure can induce physical and mental disorders in offspring, yet the effect of paternal ethanol exposure on offspring is unclear. Methylation alterations in imprinted genes may be related to the well-documented teratogenic effects of ethanol. Here, we report that ethanol (0, 1.1, 3.3 g/kg) was administered intragastrically to male mice and a behavioral study was performed on their F1 generation. Data show that F1 mice with fathers exposed to the highest dose of ethanol had delayed cognitive performance and increased anxiety and depression. A specific circling behavior was observed in the offspring of the paternally ethanol-exposed group. The degree of methylation and mRNA expression of H19, Peg3, Ndn and Snrpn were assessed in paternal sperm and in the cerebral cortices of each offspring. It did affect methylation in paternal sperm (H19 and Peg3) and in the offspring's cerebral cortices (CpG7 and CpG11 in Peg3 and Snrpn), but the level of mRNA expression has not changed. In the circling mice, the highest ethanol exposure increase in methylation (CpG 1, 2, 7 and 11) and decreases in mRNA of Peg3.Thus, chronic paternal ethanol exposure can affect the methylation of imprinted genes in sire sperm that may be passed on to offspring, giving rise to mental deficits.• We discovered behavioral abnormities in offspring of male parents exposed to ethanol. • Aberrant methylation was observed in the DNA methylation regions of imprinted genes in paternal sperm. • The same methylation and expression changes were also observed in the offsprings' cerebral cortices. • A possible explanation for the chronic ethanol-induced paternal germline epigenetic effects on offspring development.

摘要:This paper proposes a novel waverider generated from axisymmetric supersonic flows past a pointed von Karman ogive. First, a pointed von Karman ogive is obtained by pointing the blunt nose of the original von Karman ogive. Then, the axisymmetric supersonic flow field past the pointed von Karman ogive is calculated using the method of characteristics, and this calculated flow field is employed as the new basic flow field of a waverider. A novel waverider is generated from this new basic flow field by a streamline tracing technique that is based on left-running Mach lines. Second, numerical methods are employed to validate the applicability of this new design concept to an aerodynamic configuration. Finally, a comparison model is designed using the conventional design concept and its performance is numerically predicted to analyze the differences between the new and conventional design concepts. The comparison results show that the novel waverider possesses higher lift-to-drag ratios, smaller trim drag, and larger internal volume than the conventional waverider; the results also demonstrate that the proposed design concept can provide greater flexibility in the design and optimization of hypersonic waverider vehicles.

13、Novel Osculating Flowfield Methodology for Hypersonic Waverider Vehicles Based on Variable Shock Angle

摘要:This study proposes a variable shock angle osculating flowfield waverider design method. The flow field structure and aerodynamic characteristics of osculating flowfield waveriders are also analyzed. In this method, the constraint of imposing the same conical flowfield on each osculating plane is removed because the shock angle of each osculating plane is different, which can be chosen according to the specific needs of waveriders. The obtained results show that the shock angle distribution is an important factor in the design of osculating flowfield waveriders. A waverider whose shock angle increases as a parabolic curve along the spanwise direction has a larger volume and lower lift-to-drag ratio. In contrast, a waverider whose shock angle decreases as a parabolic curve along the spanwise direction has a smaller volume and higher lift-to-drag ratio. Providing different shock angle distributions can lead to greater flexibility in the design and optimization of hypersonic waverider vehicles.

14、Small ubiquitin-related modifier 2/3 interacts with p65 and stabilizes it in the cytoplasm in HBV-associated hepatocellular carcinoma

摘要:Abstract Background SUMOylation, an important post-translational modification, associates with the development of hepatocellular carcinoma (HCC). p65, one of the most important subunits of NF-κB, is a key regulator in the development of HCC and has been reported to be SUMOylated by exogenous small ubiquitin-related modifier 3 (SUMO3) in HEK 293T cells. However, the relationship between p65 and SUMO2/3 in HCC remains unknown. This study was to investigate the interaction between p65 and SUMO2/3 and explore the potential roles involved in HCC. Methods The expressions of p65 and SUMO2/3 in the liver tissues were detected by using immunohistochemistry. We performed double-labeled immunofluorescence and co-immunoprecipitation assay to verify the interaction between p65 and SUMO2/3. The extraction of nuclear and cytoplasmic proteins was performed, and the subcellular localization of p65 was detected. The proliferation and migration of hepatoma cells were observed using MTT, colony formation, and transwell assays. Results We found a strong SUMO2/3-positive immunoreactivity in the cytoplasm in the non-tumor tissues of HCC. However, SUMO2/3 level was down regulated in the tumor tissues as compared with the adjacent non-tumor tissues. In accordance with this finding, p65 was up regulated in the adjacent non-tumor tissues and almost localized in the cytoplasm. There was a close correlation between SUMO2/3 and p65 expressions in the liver tissues (R = 0.800, p = 0.006). The interaction between p65 and SUMO2/3 was verified by co-immunoprecipitation and double-labeled immunofluorescent assays. TNF-α (10 ng/ml) treatment for 30 min not only up regulated the cytoplasmic conjugated SUMO2/3, but also enhanced SUMO2/3-p65 interaction. Furthermore, we found that SUMO2/3 up regulated the cytoplasmic p65 protein level in a dose-dependent manner, but not affected its mRNA level. The increase of p65 protein by SUMO2/3 was abolished by MG132 treatment, a reversible inhibitor of proteasome. Meanwhile, TNF-α-induced increase of SUMO2/3-conjugated p65 was along with the reduction of the ubiquitin-conjugated p65. The further study showed that SUMO2/3 over-expression decreased the proliferative ability of hepatoma cells, but did not affect the migration. Conclusion SUMO2/3-p65 interaction may be a novel mechanism involved in the transformation from chronic hepatitis B to HCC via stabilizing cytoplasmic p65, which might shed light on understanding the tumorigenesis and development.

摘要:We proposed a two-coupled microsphere resonator structure as the element of angular velocity sensing under the Sagnac effect. We analyzed the theoretical model of the two coupled microspheres, and derived the coupled-resonator-induced transparency (CRIT) transfer function, the effective phase shift, and the group delay. Experiments were also carried out to demonstrate the CRIT phenomenon in the two-coupled microsphere resonator structure. We calculated that the group index of the two-coupled sphere reaches n g = 180.46, while the input light at a wavelength of 1550 nm.

16、Facile synthesis of amorphous FeOOH/MnO2 composites as screen-printed electrode materials for all-printed solid-state flexible supercapacitor

摘要:More convenience and intelligence life lead by flexible/wearable electronics requires innovation and hommization of power sources. Here, amorphous FeOOH/MnO2 composite as screen-printed electrode materials for supercapacitors (SCs) is synthesized by a facile method, and solid-state flexible SCs with aesthetic design are fabricated by fully screen-printed process on different substrates, including PET, paper and textile. The amorphous FeOOH/MnO2 composite shows a high specific capacitance and a good rate capability (350.2 F g−1 at a current density of 0.5 A g−1 and 159.5 F g−1 at 20 A g−1). It also possesses 95.6% capacitance retention even after 10 000 cycles. Moreover, the all-printed solid-state flexible SC device exhibits a high area specific capacitance of 5.7 mF cm−2 and 80% capacitance retention even after 2000 cycles. It also shows high mechanical flexibility. Simultaneously, these printed SCs on different substrates in series are capable to light up a 1.9 V yellow light emitting diode (LED), even after bending and stretching.Display Omitted• The amorphous FeOOH/MnO2 composites are prepared by a facile method. • This material shows 350.2 F g−1 and 95.6% capacitance retention after 104 cycles. • All-printed flexible supercapacitors are fabricated on PET, paper and textile. • The device is capable to light up a 1.9 V LED, even after bending and stretching.

19、Effects of package on the performance of MEMS piezoresistive accelerometer

摘要:Package is one of the key technologies for MEMS accelerometers. This paper investigated the influence of package on performances of custom-designed MEMS piezoresistive accelerometer. Based on the designed package process, the effect of die adhesive and potting materials on the sensor sensitivity and residual stress were studied by theory analysis and experimental test. The results showed that: with more quantity of die adhesive when the thickness was fixed, the sensitivity of MEMS piezoresistive accelerometer was much smaller; in addition, with thicker die adhesive, the accelerometer had much bigger sensitivity and much smaller residual stress increment after the die attach process. Meanwhile, the sensitivity was proportional to the density of potting material, and it varied with the elastic modulus.
