129、Metabolic reprogramming by HIF-1 activation enhances survivability of human adipose-derived stem cells in ischaemic microenvironment

摘要:Objectives: Poor cell survival severely limits the beneficial effect of adipose-derived stem cell (ADSC)-based therapy for disease treatment and tissue regeneration, which might be caused by the attenuated level of hypoxia-inducible factor-1 (HIF-1) in these cells after having been cultured in 21% ambient oxygen in vitro for weeks. In this study, we explored the role of pre-incubation in dimethyloxalylglycine (DMOG, HIF-1 activator) in the survivability of human ADSCs in a simulated ischaemic microenvironment in vitro and in vivo. The underlying mechanism and angiogenesis were also studied.Materials and methods: Survivability of ADSCs was determined in a simulated ischaemic model in vitro and a nude mouse model in vivo. Cell metabolism and angiogenesis were investigated by tube formation assay, flow cytometry, fluorescence staining and real-time polymerase chain reaction (RT-PCR) after DMOG treatment.Results: The results of the experimental groups showed significant enhancement of ADSC survivability in a simulated ischaemic microenvironment in vitro and transplanted model in vivo. Study of the underlying mechanisms suggested that the improved cell survival was regulated by HIF-1-induced metabolic reprogramming including decreased reactive oxygen species, increased intracellular pH, enhanced glucose uptake and increased glycogen synthesis. Tube formation assay revealed higher angiogenic ability in the DMOG-treated group than that in control group.Conclusions: The promotion of HIF-1 level in ADSCs induced by DMOG preconditioning suggests a potential strategy for improving the outcome of cell therapy due to increased survival and angiogenic ability.

摘要:• Novel mesoporous surface ion-imprinted polymer was prepared by RAFT polymerization. • High selectivity and satisfied adsorption capacity for Ce(III) were obtained. • Ce(III)-IIP could be used as a potential adsorbent for Ce(III) removal in wastewater. • The dynamic column performance was firstly studied to adsorb Ce(III) by Ce(III)-IIP.A novel Ce(III) ion-imprinted polymer (Ce(III)-IIP) has been prepared by surface imprinting technique with reversible addition-fragmentation chain transfer (RAFT) polymerization based on support matrix of SBA-15. The prepared adsorbent is characterized by FT-IR, XRD, SEM, TEM, nitrogen adsorption-desorption, GPC, and TGA. The results suggest that the surface imprinted polymer synthesized by RAFT is a thin layer. For adsorption experiments, Ce(III)-IIP is investigated to remove Ce(III) by column study at different flow rates, initial metal ion concentrations, and adsorption temperature. The dynamic kinetics analyses reveal that the overall adsorption process is successfully fitted with the pseudo-first-order kinetic model and the equilibrium time was 60 min. Meanwhile, the experimental data is in good agreement with Thomas model. Ce(III)-IIP has the excellent selectivity and regenerate property. Meanwhile, the proposed method has been successfully applied in the removal of Ce(III) in natural water samples with satisfactory results. All the results suggest that Ce(III)-IIP could be used as an excellent adsorbent for efficient removal of Ce(III) from aqueous solution.

摘要:Inbred strains of mice have served as valuable models for studying genetic susceptibility to drug addiction, an alternative to genetically modified mouse models. This is the first study comparing amphetamine (AMPH) effects on locomotor stimulation and dopamine efflux between two inbred strains of mice C57BL/6J and 129S2/SvHsd, frequently used as background strains for production of genetically engineered mice. There were no significant differences in basal locomotor activity and basal dopamine levels between the two strains. However, C57BL/6J mice showed greater AMPH-stimulated locomotor activity and AMPH-induced striatal dopamine efflux than 129S2/SvHsd mice. The differential AMPH effects could not be explained by differences in presynaptic dopamine components such as surface and total dopamine transporter (DAT) expression levels, striatal dopamine contents, and DAT activity. C57BL/6J and 129S2/SvHsd mice are excellent models for future identification of genetic, molecular, and behavioral components related to individual vulnerability to AMPH addiction. This study emphasizes the importance of mouse strain selections in the production of genetically modified mice for investigating phenotypes and mechanisms of psychostimulants.

137、Two different restriction-modification systems for degrading exogenous DNA in Paenibacillus polymyxa

摘要:Accompanied by benefits from horizontally transferred genes, bacteria have to face the risk of the invasion of dangerous genes. Bacteria often use the restriction-modification (R-M) system, which is consisted of methyl transferase (MEase) and restrictase (REase), to protect self-DNA and defend against foreign DNA. Paenibacillus polymyxa, widely used as growth promoting rhizobacteria in agriculture, can also produce compounds of medical and industrial interests. It is unclear whether R-M systems exist in P. polymyxa. In this study, we used a shuttle plasmid with epigenetic modification from different bacteria to explore R-M systems in P. polymyxa. We found that DNA which is methylated by DNA adenine methyltransferase (Dam) in E. coli was strongly restricted, indicating the presence of a Dam-methylation-dependent R-M system in P. polymyxa. 摘要:Financial factors have been found highly important in influencing firms』 real activities and in promoting aggregate growth. Yet, the linkage between finance and firm-level productivity has been overlooked in the literature. We fill this gap using a panel of 130,840 Chinese manufacturing firms over the period 2001-2007 to estimate a TFP model augmented with cash flow. We find that, especially for illiquid foreign and private firms, productivity is strongly constrained by the availability of internal finance. Furthermore, contrary to private firms, foreign non-exporters display higher dependence of productivity on cash flow than exporters.
